Genetic Determinants of Rifampicin Resistance of Mycobacterium tuberculosis not Included in the Russian Molecular Genetic Test Systems

The objective: to study the genetic determinants of rifampicin resistance Mycobacterium tuberculosis by sequencing RRDR rpoB gene. Subjects and Methods. included 651 M. isolates that were tested for drug suceptibility (DST) genotypic methods (gDST) TB-TEST (BIOCHIP-IMB, Russia), Amplitube-MDR-RV (NPK Sintol, Russia) phenotypic (phDST): absolute concentration method, BACTEC MGIT 960 system, Sensititre Myco TB kit. Sanger gene was performed 20 with discrepancies in results phDST gDST. Results. As a result sequencing, two variants mutations region found, which not detected Amblitub MDR-RV test systems, however strongly associated tuberculosis. most common variant insertion three nucleotides (TTC) encoding amino acid phenylalanine at position 434 codon (1296-1300insTTC).
